Effect of Sequential Mestranol-Chlormadinone on Sebum Production

Abstract

The effect on sebum production from the cyclic administration of an oral sequential estrogen-progestin drug was studied in 14 women, ages 18 to 24 years, three of whom had acne. The drug, which was administered for 11 to 92 weeks, consisted of tablets containing 0.08 mg of mestranol for the first 11 days and tablets containing 0.08 mg of mestranol plus 2 mg of chlormadinone acetate for ten days. Eight subjects received two tablets daily, and six subjects were given one tablet daily. A significant degree of sebaceous gland suppression occurred in ten subjects including the patients with acne. Once sebaceous gland activity had been reduced, the acne cleared. Of five subjects who received prolonged treatment, 31 to 92 weeks, one demonstrated a return of sebum levels toward normal in the tenth month despite continuing drug administration.
