KAPOSI SARCOMAASSOCIATED human herpesvirus, or human herpesvirus 8 (HHV-8), DNA sequences were first identified in 1994 using representational difference analysis on Kaposi sarcoma (KS) and healthy tissue from a patient seropositive for the human immunodeficiency virus (HIV).1 Until now, HHV-8 has been definitively associated with 3 human diseases: KS, primary effusion lymphoma (PEL) (also called body cavitybased lymphoma),2 and multicentric Castleman disease.3
In contrast, the link between HHV-8 and myeloma and sarcoidosis remains controversial.4-5 From 1 PEL-derived cell line, a 140.5-kilobase genome of this herpesvirus was sequenced and 81 open reading frames were demonstrated.6 Strong homologous characteristics with herpesvirus saimiri and, to a lesser extent, Epstein-Barr virus were evident. Human herpesvirus 8 encodes for structural proteins and enzymes common to most herpesviruses but also for unspliced genes homologous to cellular genes that probably have been captured from the host cells during viral evolution.
